RESUMO

OBJECTIVE@#To investigate the molecular mechanism by which miR-20a-5p regulates HOXB13 gene expression and inhibits lung cancer cell proliferation.@*METHODS@#The expression levels of HOXB13 mRNA and protein in lung cancer A549 cells transfected with HOXB13 overexpression plasmid or HOXB13 siRNA were detected with real-time fluorescence quantitative PCR (qRT-PCR) and Western blotting. CCK-8 and EdU assays were used to examine the effect of modulation of HOXB13 expression on cell proliferation. We screened possible binding miRNAs of HOXB13 by bioinformatics analysis. In A549 cells transfected with miR-20a-5p mimic or miR-20a-5p inhibitor, the expression level of miR-20a-5p was detected by qRT-PCR and the protein expression of HOXB13 was determined with Western blotting. CCK-8 and EdU assays were used to assess the effect of miR-20a-5p overexpression on the proliferation of A549 cells. miR-20a-5p mimic and HOXB13 overexpression plasmids were co-transfected into A549 cells, and the changes in cell proliferation were evaluated with CCK-8 and EdU assays.@*RESULTS@#HOXB13 overexpression obviously promoted the proliferation of A549 cells (P < 0.05). miR-20a-5p was identified as the potential binding miRNA of HOXB13. Overexpression of miR-20a-5p in A549 cells significantly decreased the expression of HOXB13 protein (P < 0.05), while interference of miR-20a-5p obviously increased HOXB13 expression (P < 0.05). The results of cell proliferation experiment showed that miR-20a-5p and HOXB13 had opposite effects on cell proliferation, and the cells overexpressing both miR-20a-5p and HOXB13 showed a lower proliferation activity than the cells overexpressing HOXB13 but higher than the cells overexpressing miR-20a-5p alone (P < 0.05).@*CONCLUSION@#miR-20a-5p inhibits proliferation of lung cancer cells by down-regulating the expression of HOXB13.

Objective To observe the effect of β-carotene ( β-C ) on the learning and memory and the expression of caspase-3 and p-tau expression in hippocampus of obstructive sleep apnea syndrome ( OSAS) rats. Methods Twenty-four adult SD rats were randomly divided into normal control group, OSAS model group ( model group) and β-C intervention group (intervention group), with 8 rats in each group. A hypoxic chamber was used to establish an OSAS rat animal model. The rats in model group and intervention group were given by gavage before intragastric administration, and no treatment was performed in the normal group. After completion of the modeling, the Morris water maze was used to test the learning and memory ability of the experimental rats. Observing the pathological changes of hippocampal tissue in rats by HE staining. The protein expression of caspase-3 and p-tau in hippocampus was detected by Western Blotting. Results Escape latency time of model group in each time point were significantly prolonged than those in normal control group ( all P<0. 05), and escape latency time of intervention group at 2-5 d were significantly shorter than those in model group (all P<0. 05). The number of times of crossing the platform in model group was significantly less than that in normal control group and intervention group ( all P<0. 05). Compared with those in model group, the expression of caspase-3 and p-tau in normal control group and intervention group were significantly lower (all P<0. 05). Conclusion β-C can reduce the impairment of learning and memory ability caused by OSAS, and the mechanism may be related to its inhibition of caspase-3 and p-tau protein expression.

RESUMO

AIM: To evaluate the efficacy and safety of vitrectomy combined with internal limiting membrane ( ILM) peeling for treating foveoschisis in high myopia. METHODS:Thirty high-myopia patients (30 eyes) with foveoschisis from March 2011 to March 2013 were divided two groups: the treatment group ( 16 eyes ) was treated with vitrectomy combined with ILM peeling, and the control group (14 eyes) was treated only with vitrectomy. The foveoschisis reattachment and and best spectacle-correction were measured preoperatively and 2mo after surgery. RESULTS: The improvement of foveoschisis reattachment and best spectacle - correction in the treatment group was significantly better than those in the control group (P CONCLUSION: Vitrectomy combined with ILM peeling is a safe and effective treatment for foveoschisis in high myopia.

<p><b>OBJECTIVE</b>To investigate the late results of using posterior restoration and three-column fixation to treat lumbar spondylolisthesis.</p><p><b>METHODS</b>One hundred and eighty-four patients with lumbar spondylolisthesis were collected from March 1999 to May 2007, they were treated by posterior restoration and fixation with single nail-grooved tail steel plate and fixed with cage (WDFC). Among these cases, 87 cases were fixed with one WDFC, 97 cases were used two WDFCs.</p><p><b>RESULTS</b>All patients were followed up for 8 to 69 months(averaged 23 months). According to Nakai standard, the results was excellent in 142 cases, good in 34, fair in 8, the excellent and good rates were 95.6%. Seventy-nine vertebraes with I degree spondylolisthesis were reduced after surgery. Eighty-seven vertebraes with II degree spondylolisthesis were reduced except 9 with I degree spondylolisthesis left. Twenty-one with III degree spondylolisthesis were reduced except 5 with I degree spondylolisthesis left; In 2 with IV degree spondylolisthesis, one with I degree spondylolisthesis left and the other with II degree spondylolisthesis left. The follow-up results showed that there was no statistical significance in the height of intervertebral space between preoperation and post-operation, and no recurrence was observed and no single nail-grooved tail steel plate and WDFC were loose or crashed.</p><p><b>CONCLUSION</b>Posterior restoration and three-column fixation is a positive modus operandi to treat lumbar spondylolisthesis,which can reduce excellently,keep the height of intervertebral space and stabilization of segment, obtain high rate of fusion, and cut down complication.</p>

Objective To evaluate the efficacy of the treatment of hilar biliary malignant obstruc tion with multiple stent drainage.Methods One hundred and twenty-seven patients with malignant biliary hilar obstruction were enrolled.The obstructions at the common hepatic duct within less than 1 cm to the junction of the left and the right hepatic duct were found in 66 cases,at the proximal common hepatic duct and the left and the right hepatic ducts in 45 cases,at the right hepatic duct in 5 cases and at the both left and right hepatic duets in 11 cases.Sixty-six patients received stent placement through the right biliary ducts and the common bile duct by puncturing the right mid-axillary line.The other 37 patients received 2 stents placement (disposed"Y"style) through the left and the right hepatic duct punc turing routway.Seven patients received 2 stents placement (disposed"┌"style) through the right hepatic duct and the common bile duct with a stent placed between the left and the right hepatic duct.Three patients had right hepatic duct stent placed first,followed by right hepatic duct and common hepatic duct stent. Twelve patients had stents placed in the right hepatic duct with external drainage from the left hepatic duct. Two patients had multiple strictures at the right hepatic duct,who got multiple external drainages.The total serum bilirubin levels were measured pre-and post-operatively.Results One hundred and twenty-seven patients with bi[iary obstraction had internal stents placed for drainage.The average total bilirubin levels among 121 patients were (283.4?175.4 )?mol/L pre-operation and (63.2?11.8)?mol/L post-operation (P
